Shikonin Induces Oxidative Damage and Promotes Cell Senescence in Lung Cancer Cells through p53/p21WAF Signaling Pathway

To investigate the effect of shikonin on oxidative damage lung cancer cells and promotes cell senescence through p53/p21Waf signaling pathway. The adenocarcinoma A549 were cultured made into suspension. for 12 h, 24 h 48 respectively with different concentrations (0 μm, 2 μm 4 μm) activity was detected by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide method. cycle each group measured flow cytometry after action shikonin. aging determined beta-galactosidase staining reactive oxygen species content in expression levels cyclin D1, demethylase KDM2B, deoxyribonucleic acid marker protein p-H2AX, p53 p21WAF Western blot. With increase concentration, D1 KDM2B significantly decreased, proportion resting phase/intermediate phase senescent increased, level increased (p<0.05). Shikonin can inhibit activity, induce promote senescence, which may be achieved activating p53/p21WAF
